AATCC-100 is a quantitative capacity analysis method used for evaluating the antibacterial rate of antimicrobial textiles.

This method was proposed by the AATCC Committee in 1961 and revised in 1965, 1981, 1988, and 1993. It is currently one of the widely used antibacterial testing methods abroad.

The principle of the method is as follows: test bacteria are inoculated onto the sample to be tested and a control sample, a certain amount of neutralizing solution is added, and the bacteria are washed off by vigorous shaking. The bacterial concentration in the eluate is measured using the dilution plate method, and the percentage reduction of bacteria on the fabric is calculated compared to the control sample. The drawbacks of this method are that it cannot test too many samples in one experiment, it is time-consuming; it cannot evaluate the antibacterial performance of non-leaching samples; it does not specify the composition of the neutralizing solution in detail; the nutrient content in the bacterial solution is too rich, which differs significantly from actual wearing conditions; and the container is too large, making it difficult to handle [13].

Based on domestic and international experience and through extensive experimentation, the method has been improved to form a systematic quantitative testing methodology that can meet the needs of testing the antibacterial performance of various antimicrobial textiles. This improved version, known as the modified AATCC-100, includes the following key points:

The sample size of the AATCC-100 method is changed from a circular shape with a diameter of 4.8 cm to a square with a side length of approximately 1.8 cm, and it is placed in a 30 mL or 50 mL capped conical flask. AATCC broth is replaced with 0.85% ice-cold physiological saline (0-4°C) to dilute the inoculum. The bacterial concentration is diluted from about 10^8-10^9 CFU/mL to 1×10^5-2×10^5 CFU/mL to prepare the inoculum. The neutralizing agent is replaced with 20 mL of 0.85% ice-cold physiological saline to wash the sample [14].

The antibacterial activity and bactericidal activity of the sample are calculated using the following formulas:

Antibacterial Rate = (Viable bacteria count of blank control after 18h - Viable bacteria count of sample after 18h) / Viable bacteria count of blank control after 18h × 100%

Bactericidal Rate = (Viable bacteria count of blank control at '0' time - Viable bacteria count of sample after 18h) / Viable bacteria count of blank control at '0' time × 100%

This method can be used for both leaching and non-leaching samples, and the nutrient content of the medium is suitable for the conditions of fabric use [12].
