When using an oil immersion lens to observe bacteria, it is often difficult to locate the object image. This is a common issue for first-time microscope users, and many students encounter this problem. There are three main reasons: first, the oil immersion lens may be contaminated; second, the field of view may be too dark; third, the fine adjustment knob may have been turned too quickly, raising the barrel too fast. The solution is to first clean the oil immersion lens with lens paper dipped in a small amount of xylene to ensure it is clean. Then adjust the light to brighten the field of view. While watching from the side, slowly lower the barrel so that the oil immersion lens is immersed in cedarwood oil, almost touching but not pressing against the slide. Then slowly turn the fine adjustment knob to raise the barrel. During this process, the object image should become visible. If not, repeat the above steps until the image is seen.

Common issues and solutions when cleaning high-power or oil immersion lenses?

When cleaning high-power lenses (40x objective) that are contaminated or oil immersion lenses (100x objective) after use, many students use too much xylene. This not only wastes xylene but also risks dissolving the adhesive on the objective lens, causing the lens to detach. The wiping motion should be gentle, without applying excessive force.
